Luciferase Test | GFP-Test | Typical Calibration Curve
 

Luciferase Test

Assay

  • Thaw frozen stocks of biosensor cells and revive them in sterile luria broth
  • Mix biosensor suspensions with the aqueous samples directly in luminometer titer plates
  • Tape plates with Parafilm and incubate them in a rotary shaker at 30°C for at least 30 minutes
  • Add a drop of decanal solution to the assay mixtures
  • Mix and incubate for another 3 minutes
  • Measure light emission by luminometry

Material

  • biosensor strain Escherichia coli (pJAMA-arsR)
  • luminometer
  • luria broth
  • arsenite stock solution
  • thermomixer
  • n-Decanal solution

Data handling

Arsenic concentrations in the samples are interpolated from a calibration curve.

  • Tape plates with Parafilm and incubate them in a rotary shaker at 37°C for at least 30 minutes
  • Measure light emission by fluorimetry

  • fluorimeter
  • MOPS 3-[N-Morpholino] propanesulfonic acid
  • arsenite stock solution
  • thermomixer

Data handling

Arsenic concentrations in the samples are interpolated from a calibration curve.
