Recombinant AAV vectors of various serotypes (1,2,3,4,5,8) are produced by co-transfection of monolayers of HEK-293T cells and purified according a well-established method (Zolotukhin et al., 2002; Methods. 28:158-67)
Viral stocks are titrated using methods to determine capsids, viral genomes and infectious titers (Tenenbaum et al., 1999; Lock et al., 2011).
