- By using in-house ES cells, the generation of chimeras and germ line transmission should occur in the vast majority of cases
Delivery of a minimum of 3 chimeras
Potential drawbacks:
- lack of homologous recombination in ES cells might indicate problems in the DNA construct or that gene targeting interferes with mouse development
Injection of targeted ES cell clones

Potential drawbacks:
Use of ES cells from outside (e.g., from other groups, companies) may influence the degree of chimerism and can subsequently influence the potency to confer germline transmission. Thus, germline transmission is likely but cannot be guaranteed
