Electroporation of targeting vectors into cells

(including electroporation, selection of 500-700 ES cell clones, recovery, amplification and freezing of clonal stocks)

Gene Targeting

 CHF 18'000.-

(including electroporation, selection of ES cells, recovery, amplification and freezing of clonal stocks, pre-testing of ES cells before injection (chromosome number, mycoplasma testing), embryo collection, injection of at least 2 targeted ES cell clones into blastocysts, transfer into pseudopregnant recipients, keeping offspring up to weaning)

- By using in-house ES cells, the generation of chimeras and germ line transmission should occur in the vast majority of cases

 

Delivery of a minimum of 3 chimeras

 

 

Potential drawbacks:

  • lack of homologous recombination in ES cells might indicate problems in the DNA construct or that gene targeting interferes with mouse development

Injection of targeted ES cell clones

 CHF 6'000.-

(including amplification and freezing of clonal stocks, pre-testing of ES cells before injection [chromosome number, mycoplasma testing], embryo collection, injection of at least 2 targeted ES cell clones into blastocysts, transfer into pseudopregnant recipients, keeping offspring up to weaning)

 

Potential drawbacks:

Use of ES cells from outside (e.g., from other groups, companies) may influence the degree of chimerism and can subsequently influence the potency to confer germline transmission. Thus, germline transmission is likely but cannot be guaranteed
