Title: "High-resolution motion-correction in Magnetic Resonance Imaging"

Outline: As MR images are pushed to higher spatial resolutions, so they become more sensitive to unavoidable small movements of the subject, even in healthy volunteers. I will present initial results using highly-accelerated fat images acquired interleaved with the main acquisition in order to track, and correct for, these small movements. Image quality is noticeably improved, even for very small head movements.

Title: "Challenges and Developments in Multimodal Quantitative Computational Anatomy"

Outline: In this talk, I shall outline previous advances with multi-parameter mapping and advanced quantitative segmentation of the human brainstem and cortex. I will discuss the problems pathological brain tissue poses to current methodological frameworks that have been developed using healthy controls, and highlight these issues using a longitudinal dataset of patients with small vessel disease. Additionally, I will introduce recent collaberative work examining “anomalous diffusion” (aDTI), a technique that utilises a continuous random time walk model to accurately parameterise the diffusion curve, providing insights into tissue microstructural properties beyond fractional anisotropy. I will demonstrate some of the biophysical properties of the quantitative aDTI parameters using the Human Connectome Project dataset. Finally, I will use a variety of metrics to discuss one of the problems associated with multimodal segmentation, namely the variability in clustering solutions that can be extracted from a single structure, and outline on-going work attempting to reconcile these observed differences.

"R2* plays an important role in the quantitative evaluation of the brain function and tissue iron content. Unfortunately, susceptibility induced macroscopic field inhomogeneities (B0 macro) across an image voxel act to increase the R2* in a gradient echo image. If these B0macro are measurable their influence can be removed in post processing. Conventionally, these algorithms assume the phase evolves linearly with time; however, in the presence of a large B0macro, e.g. near the edge of the brain, this is assumption is broken. The phase evolution appears random. herein, we hypothesize that the phase evolution in the presence of large B0macro, can be modeled and corrected using 1D-random-walk theory. "

"Postnatal development of excitatory transmission in the VTA."

"Postnatal periods of development are characterized by rapid changes in neuronal networks, providing critical periods in which environmental experience can lead to long-term changes in brain and behaviour. In the Ventral Tegmental Area (VTA), postnatal maturation of excitatory transmission is regulated by mGluR1, which orchestrates both the subunit composition of AMPARs and NMDARs. We investigated the molecular mechanisms underlying mGluR1 activation and found that selective manipulation of a gene associated with Autism Spectrum Disorders (ASDs) in the VTA leads to an incomplete postnatal maturation of excitatory transmission in the VTA suggesting that VTA may be implicated in some of the behavioural symptoms observed in ASDs."

"Restless at rest: dynamic functional connectivity as a new measure to probe brain network organization"

Functional connectivity (FC) as extracted from fMRI data has become a key measure to study the brain's functional integration. FC is obtained as the correlation between timecourses of different voxels or brain regions, conventionally computed for the full duration of the resting-state scanning session. Recently, temporal variations in FC have been observed using sliding-window correlation, for which the term dynamic functional connectivity (dynFC) has been coined. Despite the risk of spurious fluctuations due to short window length, we reported meaningful dynFC that reflected complementary information w.r.t. standard FC. We recently proposed a multivariate framework to facilitate the analysis and interpretation of dynFC in terms of FC building blocks. We also showed clinical relevance of dynFC; e.g., resting-state fluctuations of FC allow to discriminate healthy controls from minimally-disabled MS patients.

"Neural representation of time intervals in humans."

  

Precise estimation of time interval is crucial for our perception and behavior. Several theoretical models of time perception have predicted that elapsed time is explicitly represented by a population of neurons that fire selectively for their preferred durations, however, the empirical evidence has been missing. In this talk, I will describe my recent fMRI study that provides evidence of explicit duration representations in the human brain.

"Biophysical compartment diffusion models: application to the developing human brain"

 

Diffusion tensor imaging (DTI) has demonstrated its unique ability to probe cellular architecture and brain connectivity. Despite the valuable information that DTI provides about the general organization, integrity and maturation of fiber bundles, the interpretation of the changes on a cellular scale remains very challenging. Recently, new biophysical compartment diffusion model, based on the local diffusion process (such as restriction, hindrance or free diffusion), have been proposed, which make possible to relate diffusion-weighted signals directly to the cellular structure. I will present you the basis of these new biophysical diffusion models and the application of CHARMED and NODDI to the developing human brain.

 "Cognitive Consilience - the blueprint of cognition through the 7 neuroanatomical circuits underlying cognitive information processing."

 

Cognitive Consilience is an attempt to form a comprehensive and cohesive understanding of the primate non-primary neuroanatomical cognitive circuitry through consilience (the integration of knowledge). The first goal is to assemble and synthesize a comprehensive accurate neuroanatomical picture correlated to basic psychological function (eg working memory, short-term memory, etc), fully referenced from hundreds of original neuroanatomy studies. Secondly we utilize web and iPad technology to produce an interactive visualization that can be explored dynamically. The talk will provide the motivation for such a daunting effort, present new integrated neuroanatomical facts with hypothesis on brain function, walk through the interactive website, and discuss the future of information integration to form comprehensive theories in science in the modern age.
